Papain

A cysteine protease from papaya fruit that cleaves proteins to support smooth digestion and gastrointestinal comfort.

What it is

Papain is a cysteine endopeptidase derived from the latex of unripe papaya fruit (Carica papaya). It contains a reactive sulfhydryl group that hydrolyzes peptide bonds, amides, and esters across a wide pH range (4.5 to 8.5) and at temperatures up to 60°C. In dietary supplements, standardized papain powder (expressed in USP units or PU) effectively digests animal and plant proteins in both gastric and duodenal environments.

Papain acts as an upstream enzymatic bio-accelerator by breaking complex proteins down into small oligopeptides and bioavailable amino acids. These cleaved protein fragments provide essential nitrogenous nutrients that support the growth and metabolic colonization of beneficial Bifidobacterium and Lactobacillus strains in the intestinal tract. In turn, probiotic microbes ferment unabsorbed peptide residues into beneficial bioactive metabolites while reinforcing the gut epithelial barrier through tight junction protein upregulation. Probiotics also maintain a balanced, slightly acidic luminal pH, optimizing endogenous protease stability, inhibiting putrefactive bacterial overgrowth, and mitigating gut inflammation.
